چکیده

Finely divided thorium and carbon were injected into the rat lung via the trachea and their uptake was studied electron microscopically. Small amounts of thorium were found in the lamellar vacuoles of the granular pneumocytes. Their presence was maximal at 24 hours and they later disappeared from these cells. This phenomenon is thought to represent a passive inflow as the vacuoles open on to the alveolar surface to discharge their content of surfactant, rather than evidence that the granular pneumocyte is concerned in surfactant disposal. An explanation reconciling the secretory nature of the vacuoles with their previously demonstrated content of lys, osomal enzymes is advanced. Small quantities of thorium dioxide were ingested by the

Metabolism of verapamil in cultures of rat alveolar epithelial cells and pharmacokinetics after administration by intravenous and inhalation routes.

Administration of therapeutic entities by inhalation opens new possibilities for drug entry into systemic circulation, but this requires passage through the alveolar epithelium. Little is known about the pulmonary metabolism of verapamil. Specifically, this cardiovascular drug suffers from extensive first pass metabolism. We therefore evaluated the metabolism of verapamil in cultured alveolar e...

Pulmonary surfactant protein D in first-line innate defence against influenza A virus infections.

Influenza A viruses (IAV) cause respiratory tract infections annually associated with excess mortality and morbidity. Nonspecific, innate immune mechanisms play a key role in protection against viral invasion at early stages of infection. A soluble protein present in mucosal secretions of the lung, surfactant protein D (SP-D), is an important component of this initial barrier that helps to prev...

The genetic approach in pulmonary fibrosis: can it provide clues to this complex disease?

Multiple investigators have undertaken genetic studies in idiopathic pulmonary fibrosis populations in attempts to define genetic links to disease in hopes that this would improve understanding of disease pathogenesis and target pathways for therapy. Multiple genes have been evaluated using a candidate gene approach with limited success, with results suggesting a disease modifier effect rather ...

Tobacco smoking and the pulmonary surfactant system.

To explain enhanced retention of water insoluble gases and water insoluble particulate matter (tar)when smoke is inhaled we propose that these compounds interact physically with phospholipids forming the mono-molecular layer of surfactant lining the alveolar epithelium. This would account for the alteration in surface tension lowering properties of surfactant when exposed to whole smoke but not...

Binding of pulmonary surfactant proteins A and D to Aspergillus fumigatus conidia enhances phagocytosis and killing by human neutrophils and alveolar macrophages.

To determine whether the lung surfactant proteins A (SP-A) and D (SP-D) are involved in the initial protective immunity against opportunistic pulmonary fungal infections caused by Aspergillus fumigatus, we performed a series of in vitro functional studies to see if SP-A and SP-D enhanced binding, phagocytosis, activation, and killing of A. fumigatus conidia by human alveolar macrophages and cir...
